Bus is the most common way to travel in Cambodia, and that's why I'm going to focus on this.

I bought my ticket from Siem Reap to Phnom Penh through Neak Krorhorm (April 2010). $10 for a minibus ride -- efficient,
comfortable and on-time -- that's all I can say!

How and when to buy bus tickets
You can either purchase a ticket through:
A) The bus company -- track down the nearest office of the bus companies mentioned above
B) Buy bus tickets through a travel agent (the travel agents and guesthouses cooperate with the various bus companies)
C) Get your bus tickets through the guesthouse you're staying at
When should you buy bus tickets: There are several departures during the day to most cities in Cambodia. Of course, if you're
planning on traveling during holidays or right before an important festival begins, you should purchase your ticket as soon as you know
when you want to leave.
There are less departures for long-distance destinations, for example from Phnom Penh/Sihanoukville to Saigon, or from Siem Reap to
Stung Treng/Ban Lung/Sen Monorom.

Phnom Penh Duration of Journeys
The capital of Cambodia connects with all major cities and towns in Cambodia.
From Phnom Penh to Siem Reap (Siemreab),
it usually takes 6 hours by bus. Another 5-hour journey to Battambang.
To Sihanoukville (Kompong Som)
the bus ride lasts for 3-4 hours depending on which company you use.
Other connections from Phnom Penh: Kampot -- 3 hours, Koh Kong
(border town if you're border crossing to Thailand) 5-6 hours, Kampong Cham -- 2 hours,
Kratie -- 5 hours, Sen Monorom -- 10 hours, Ban Lung
-- 12 hours (2-3 hours from Stung Treng), Stung Treng -- 7 hours (you have to pass Stung Treng
before crossing the border to south Laos), Takeo -- 2 hours.

From Cambodia to Vietnam by Bus
There are buses leaving for Saigon from cities like Phnom Penh (6 hours) and Sihanoukville.
From Sihanoukville, with Sorya Transport, the bus leaves at 07.15 am (saw this time schedule at a poster
in Sihanoukville in April 2010).
From Phnom Penh (Sorya Transport), the buses leave at 06:45 am, 08:30 am, 11:30 am, 13:30 pm and 14:30 pm.

Siem Reap Duration of Journeys
The Angkor town is located near Tonle Sap Lake.
With shared taxi it takes 4 hours to get to border town of Poipet
(if crossing the border to Thailand). Otherwise it takes 8 hours by bus minus the delay.
When I traveled in Cambodia in April 2010,
between Siem Reap and Phnom Penh, the road was completely asphalt-covered compared to previous years.
Boat from Siem Reap to Battambang takes about 8-12 hours depending on the season (water level), type of boat
and the boat drivers. By taxi you can get to Battambang in 2-3 hours, and by bus it takes 4 hours.
From Siem Reap to Anlong Veng: 2,5 hours. From downtown Siem Reap to
Angkor park: 10-15 minutes
Siem Reap to Stung Treng (if going to Laos)
-- 12 hours. The last time I took this route it stopped by Phnom Penh (2007).
